面议ACQUITY UPLC BEH HILIC柱使用未键合BEH颗粒以提高对高极性分析物的保留能力,同时对于极性和可离子化物质的混合物提供相对于反相色谱的正交性的分离选择性。该色谱柱经优化并测试,以确保在UPLC HILIC条件下能提供高效、重现的分离效果。该产品在设计伊始就针对克服既有HILIC固定相的一个重要弱点:化学稳定性。结实的BEH杂化颗粒的宽pH耐受能力拓宽了HILIC应用条件范围,并显著延长了柱使用寿命。
目的:建立超高效亲水性液相色谱与串联四级杆飞行时间质谱仪联用技术(UPLC HILIC/Q-TOF-MS)对毛茛黄酮类成分进行分析的方法。方法:用ACQUITY UPLC BEH HILIC色谱柱,以甲醇∶水=1∶1含10mM乙酸铵(A)/乙腈∶甲醇=95∶5含10mM乙酸铵(B)梯度洗脱,使用ESI离子源,正、负离子模式下采集数据。结果:根据元素组成分析、二级质谱和相关文献,鉴定、推断出毛茛中11个主要成分,分别是小麦黄素,牡荆素,荭草素,异荭草素,芹菜素-6-C-β-D-葡萄糖苷-8-C-α-L-阿拉伯糖苷,小麦黄素-7-O-β-D-葡萄糖苷,芦丁等。结论:经过超高效亲水性液相色谱的分离,借助Q-TOF-MS测定的准确分子量及二级质谱信息可以判定毛茛中的主要黄酮类成分,为毛茛黄酮类成分分析提供了一种快速有效的方法。
